Protocol S2433, Randomized Phase III Study of Second-Line Chemotherapy with or without Panitumumab for KRAS Wild Type, Locally Advanced or Metastatic Pancreatic Adenocarcinoma

We are doing this study because we want to find out if this combination is better or worse than the 
usual approach for advanced pancreatic cancer without KRAS mutations. The usual approach, 
which is chemotherapy alone, is defined as the care most people get for advanced or metastatic 
pancreatic cancer.

Chemotherapy for the Treatment of Patients With Newly Diagnosed Very Low-Risk and Low Risk Fusion Negative Rhabdomyosarcoma

Rhabdomyosarcoma is a type of cancer that occurs in the soft tissues in the body. This phase III trial aims to maintain excellent outcomes in patients with very low risk rhabdomyosarcoma (VLR-RMS) while decreasing the burden of therapy using treatment with 24 weeks of vincristine and dactinomycin (VA) and examines the use of centralized molecular risk stratification in the treatment of rhabdomyosarcoma. Another aim of the study it to find out how well patients with low risk rhabdomyosarcoma (LR-RMS) respond to standard chemotherapy when patients with VLR-RMS and patients who have rhabdomyosarcoma with DNA mutations get separate treatment. Finally, this study examines the effect of therapy intensification in patients who have RMS cancer with DNA mutations to see if their outcomes can be improved.

A Phase III, Randomized, Open-Label, Multicenter, Global Study of Puxitatug Samrotecan (AZD8205) Monotherapy versus Physician's Choice of Chemotherapy in Participants with B7-H4-Selected Advanced/Metastatic Endometrial Cancer Who Progressed on or After Platinum-Based Chemotherapy and Anti-PD-1/Anti-PD-L1 Therapy (BLUESTAR-Endometrial01)

This is a Phase III, 2-arm, randomized, open label, multicenter, global study assessing the efficacy and safety of puxitatug samrotecan compared to physician's choice of chemotherapy (doxorubicin or paclitaxel) in participants with B7-H4 selected advanced/metastatic EC that progressed following platinum-based chemotherapy and anti-PD-1/anti-PD-L1 therapy.

This study aims to see if Puxi-Sam allows participants to live longer without their endometrial cancer getting worse, or simply live longer, compared to participants receiving standard of care chemotherapy. This study is also looking to see how the treatment and the endometrial cancer affect participants' quality of life.

546C-CL-0201: A Phase 1b/2 Open-label Study to Assess the Safety and Efficacy of ASP546C in Participants with CLDN18.2-expressing Locally Advanced Unresectable or Metastatic Gastroesophageal Adenocarcinoma, Pancreatic Adenocarcinoma or Other Solid Tumor Types

The primary objective of this multicohort phase 1b/2 study is to determine the RP2D of ASP546C (XNW27011) monotherapy based on the totality of efficacy, safety and PK data collected at 2 different dose levels. The secondary objectives include evaluation of the efficacy, safety, PK and immunogenicity of ASP546C monotherapy. Safety and tolerability will be assessed via AEs, SAEs, ECOG performance status, vital signs and other laboratory results.
